Hirschhorn studied medicine at the University of Vienna and received his medical degree in 1951. He then went on to specialize in medical genetics, and in 1956 he became the first professor of medical genetics in the United States. He was a professor at the University of Pennsylvania from 1956 to 1965, and then at the University of California, Los Angeles from 1965 to 1991. Hirschhorn has made numerous contributions to the field of medical genetics, including the discovery of the genetic basis of several diseases, such as cystic fibrosis, fragile X syndrome, and Duchenne muscular dystrophy. He has also been involved in the development of genetic testing and counseling for families affected by genetic disorders. Hirschhorn is the author of several books, including The Genetics of Human Populations (1975) and The Genetics of Human Diseases (1982).
